I was looking for this info all around the net and couldn't get a definite answer. So I am posting this with an obvious title so others looking for this will find it.
NO, IT WON'T LOCK BACK YOUR PHONE TO T-MO, IT WILL REMAIN SIM UNLOCKED. I did it myself, I'm on rogers network, and before doing it myself, I PMed a very nice contributor to this forum since I heard he had done it also. Both of our phones remained unlocked and operated as intended after the WM6 update.
My phone is only SIM unlocked and the T-mobile update worked perfectly.
To get the update, visit this site:
http://www.t-mobile.com/wmupgrade/
Note that you will need the IMEI number of your phone and a T-mobile phone number (I'm sure you can find a T-mobile phone number somewhere) in order for the site to allow you the download.
Also, I did not experience any particular drawbacks to the T-mobile WM6 update, phone even seems a bit faster than WM5. I guess it just works good!!
Hope this helps people to take the plunge!!

fyi - the rogers wm6 update is WAY better than the tmo version... you should check that one out too.
better yet, get the HTC wm6 update. it blows both the tmo and rogers versions out of the water.
even better... there's a cooked wm6.1 rom as well. it's not official... but by most accounts it's better than wm6.

The Rogers rom seems to have been obliviated from the surface of the web...
HTC's website doesn't have it anymore,
http://www.htc.com/member/RomList.aspx doesn't give me any rom list, maybe it's just me, if it is me, someone please tell me how dumb I am and then point me out what I am doing wrong!
At least I know where to get the cooked one..
Yeah i've also been there... www.rogers.com/s621upgrade
Found the HTC one also:
